STX-1150: The One-Shot CRISPR Cholesterol Treatment

What is STX-1150?

STX-1150 is an experimental treatment that aims to lower “bad” cholesterol (LDL) with just one single dose — and keep it low for years.

It is made by Scribe Therapeutics, a biotech company in California that just raised $129 million by going public on the stock market in July 2026.

Think of it like this: instead of taking a pill every day for the rest of your life, you get one treatment and your body does the rest.

How does it work?

Your liver has a gene called PCSK9. This gene acts like a switch that controls how much bad cholesterol stays in your blood. When the switch is “on,” more LDL cholesterol floats around, which can clog your arteries over time.

Some lucky people are born with a version of this gene that barely works. They have naturally low cholesterol and much lower rates of heart attacks — their whole lives.

STX-1150 tries to give everyone that same advantage. It uses a tool called CRISPR to gently turn down the PCSK9 switch in your liver cells.

Here is the clever part: it does NOT cut or permanently change your DNA. Instead, it puts a “mute button” on the gene — a technique called epigenetic silencing. The gene is still there. It just stays quiet. And because no DNA is permanently changed, the effect could potentially be reversed if needed.

Who is it for?

STX-1150 is being tested in adults who have high LDL cholesterol and are at higher risk for heart disease. These are people who might otherwise need daily statins or regular injections of PCSK9 inhibitor drugs (like Repatha or Praluent).

The idea is that one treatment could replace years — maybe decades — of daily pills.

Where does it stand?

The Phase 1 trial will enroll up to 64 adults. They will receive one dose and be watched closely for a full year. The trial is being run at Monash Health’s Victorian Heart Hospital in Australia, led by cardiologist Dr. Stephen Nicholls.

Australia approved the trial in May 2026. The first patients are being enrolled now.

Why does this matter?

Heart disease is the #1 killer in the world. In the United States alone, someone has a heart attack every 40 seconds.

We already have drugs that lower cholesterol — statins, PCSK9 inhibitors, ezetimibe. They work. The problem is that people have to take them forever. Many stop. Some forget. Some never start. Studies show that roughly half of people prescribed statins stop taking them within a year.

STX-1150 flips the problem on its head. Instead of asking people to remember a pill every day for 30 years, it asks for one treatment. That is a fundamentally different approach to prevention.

For longevity, this is exciting because it targets the root cause of cholesterol buildup — the PCSK9 gene itself — rather than just cleaning up the mess after it accumulates.

What we do not know yet

  • Does it work in humans? All the great results so far are from lab studies and animals. Humans are different. The Phase 1 trial will give us the first real answer.
  • Is it safe? CRISPR therapies are new. We do not have decades of safety data. The trial will watch carefully for side effects, especially anything unexpected from turning down a gene.
  • How long does it last? Scribe says “years.” We will not know the real answer until patients are followed for years. It might be 2 years. It might be 10. We simply do not know yet.
  • What happens if something goes wrong? Because this is epigenetic silencing (not permanent editing), the effect should fade over time. That is both a feature (safety) and a question (how long until you need another dose?).
  • Who will be able to afford it? Gene therapies are expensive. If STX-1150 works, the question becomes whether insurance companies will pay for it — and how much it will cost.

The bottom line

STX-1150 is one of the most exciting ideas in cardiovascular medicine: a single treatment that could lower cholesterol for years by gently silencing a gene. It just entered its first human trial, and the company behind it just went public at $129 million. There is a very long road ahead — years of testing, safety checks, and regulatory hurdles — but the science behind it is built on one of the most validated targets in heart disease research. If it works, it could change how we think about preventing the world’s biggest killer.
